A crash that claimed four lives will have a profound impact on South Australia's southeast community, police say.

Three men in their 20s and a teenager died in the single car rollover on the Riddoch Highway, near Penola, early on Saturday.

The four, aged 22, 23 and 28 and a 17-year-old boy, died at the scene. They were all local to the area.

"This is a tragedy which will have a profound effect on the families involved and the wider Limestone Coast community," Sergeant Ryan Meakin told reporters.

He urged the community to rally around the families of those killed, especially going into the festive season.

Emergency service crews who attended the scene were expected to receive counselling.

The four deaths took the state's road toll for 2016 to 83 compared to 93 at the same time last year.

The highway was closed for several hours but reopened on Saturday afternoon.
